以本项目组前期从玉米蛋白水解物中分离纯化得到的玉米蛋白单肽KSCAPLAS为研究对象,通过构建乙醇诱导LO2细胞损伤模型,探究玉米肽KSCAPLAS的拮抗乙醇性损伤作用。结果表明,KSCAPLAS能够显著降低乙醇诱导损伤的LO2细胞内活性氧(reactive oxygen species,ROS)的含量(P<0.05),并显著提高超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽还原酶(glutathione reductase,GR)活性和还原型谷胱甘肽(glutathione,GSH)水平(P<0.05),对乙醇性损伤的LO2细胞具有拮抗作用。此外,KSCAPLAS通过上调线粒体呼吸链复合物Ⅰ的含量进而上调细胞内ATP含量。与对照组相比,在肽质量浓度为100 μg/mL条件下,细胞内线粒体呼吸链复合物Ⅰ和ATP的质量分数分别提高了63.11%和31.65%。玉米肽KSCAPLAS通过促进乙醇性损伤细胞代谢提供能量,进而缓解乙醇诱导LO2细胞的损伤。本研究为玉米蛋白源功能食品的开发奠定了理论基础,为玉米源生物活性肽作用机制的研究提供新思路。 |

Taking KSCAPLAS, a single peptide of corn protein isolated and purified from the hydrolysate of corn protein, as the study object, the antagonistic effect of KSCAPLAS on ethanol induced damage of LO2 cells was explored by constructing an ethanol-induced damage model. The results showed that KSCAPLAS could significantly reduce the content of reactive oxygen species (ROS) in ethanol-induced LO2 cells (P<0.05), and significantly increase Superoxide dismutase. SOD and glutathione reductase (GR) activities and glutathione (GSH) levels (P<0.05) had antagonistic effects on LO2 cells induced by ethanol injury. In addition, KSCAPLAS up-regulates intracellular ATP content by up-regulating the content of mitochondrial respiratory chain complex I. Compared with the control group, under the condition of peptide concentration of 100 μg/mL, the mass fractions of mitochondrial respiratory chain complexⅠ and ATP were increased by 63.11% and 31.65%, respectively.
