      小米黄色素含量的定量测定不仅有助于小米产品质量评判的标准化,促进小米产品的精深加工,而且对于谷子品质育种也具有重要意义。本研究参考美国谷物化学师协会(AACC)14-50方法,研究了不同提取溶剂、提取方式、提取时间等因素对小米黄色素含量的影响,优化完善了小米黄色素含量的测定。测定谷子籽粒小米黄色素含量,以β-胡萝卜素制作标准曲线,利用水饱和正丁醇振荡提取3 h,10000 ×g 离心8 min取上清,在445 nm下定量测定小米黄色素含量;β-胡萝卜素吸光度和质量浓度在0~5.0 μg/mL范围内关系良好,相关系数r= 0.9999。该方法测定小米黄色素,相对标准偏差<3%,加标回收率92.77%~103.18%,灵敏准确、简便快速,精密度较高,适用于小米黄色素的测定。

      Quantitative determination of yellow pigment contents in foxtail millet grains in a precise manner is favorable to standardize evaluation of foxtail millet product quality, promoting further process of foxtail millet products. Moreover, it is of great significance for quality breeding for foxtail millet. The present research dealt with the improvement of the AACC method 14-50 for the determination of yellow pigment contents in foxtail millet grains through optimizing extraction solvents, methods, duration times as well as other factors. In the improved method, β-carotene was used to make a standard curve; the yellow pigment in foxtail millet grains was vibrating extracted in water saturated n-butyl alcohol for 3 h; then supernatant was retained after centrifugation at 10,000 x g for 8 min; finally, yellow pigment content was determined at 445 nm. The absorbency and contents (0 – 5.0 μg/mL) of β-carotene showed an excellent linear correlation (r2 = 0.9999). With this method, the yellow pigment content was accurately determined with the relative standard deviation < 3 % and the adding standard recovery rate ranging from 92.77 % to 103.18 %. The results demonstrated that the improved method was highly sensitive, simple and accurate for rapid quantitative determination of yellow pigment content in foxtail millet grains.
