      采用反相微乳液法,依据金属-有机框架材料一磷酸腺苷(adenosine monophosphate, AMP)&氯化锌水凝胶可以固定蛋白的原理,通过在纳米四氧化三铁磁珠表面包覆AMP&ZnCl2水凝胶并偶联抗体,制备了一种新型的水凝胶免疫磁珠。选取了玉米赤霉烯酮(Zearalenone, ZEA)单抗来制备水凝胶免疫磁珠-Fe3O4@AMP&ZnCl2?ZEA单抗磁珠,对c样品玉米粉进行了ZEA添加回收率测定,添加回收率在84.81%~90.00%之间,相对标准偏差为4.86%~9.44%;比较了Fe3O4@AMP&ZnCl2?ZEA单抗磁珠与DZTMS-PREP免疫亲和柱对6种玉米粉中 ZEA的纯化效果,测定结果和使用免疫亲和柱检测结果保持一致,说明该材料可应用于实际样品的纯化。

      A new type of hydrogel immunomagnetic beads was prepared by reverse microemulsion method. Its realization was based on the principle that the metal-organic framework AMP/ZnCl2 hydrogel can immobilize the protein, and it is wrapped AMP/ZnCl2 hydrogel on the surface of nano-ferromagnetic tetroxide beads. Zearalenone (ZEA) monoclonal antibody was selected to prepare hydrogel immunomagnetic beads-Fe3O4@ AMP ZnCl2.ZEA monoclonal antibody beads. The recovery of C sample corn flour was determined and the recoveries were between 84.61% and 90.00%, and the relative standard deviation which was 4.86% ~ 9.44%. The purification results of ZEA in six kinds of corn meal by Fe3O4@AMP ZnCl2.ZEA monoclonal antibody beads and immune affinity column were compared. The results of the assay were same as those of the immune affinity column, which indicated that the material could be used in the purification of the actual sample.
